Development of biophysical assays to study how individual enzymes function in living cells and in the test tube. Simultaneous super-resolution fluorescence imaging and single particle tracking with quantitative phase-shifted laser feedback interferometry to study how metastatic cells form and remodel their invasive structures. Imaging the formation and remodeling of the mammalian adhesion system at the bottom of cells. High-speed single particle tracking with stroboscopic illumination to study the starvation induced stringent response machinery in bacteria. In vitro single enzyme turnover assays using confocal detection. TIRF, SPR imaging and fluorescence correlation spectroscopy. Bacterial genomic integration and cloning. Protein purification, enzyme biochemistry and labeling, and oxidative folding assays.
